New Technology May Lead to Development of Cancer Therapies
Researchers at University of Delaware develop a technology to program DNA strands into switches that turn proteins on and off, according to a study conducted on March 19, 2018.
New cancer therapies and other drugs can be developed using this new technology. This study is mainly based on DNA computing, which is an emerging field in the recent years. DNA is a code that has four components including the nucleotides guanine, adenine, cytosine, and thymine. The formation of proteins depends on the arrangement of four nucleotides. The DNA strands were ordered from a manufacturer and production and purification of proteins were carried out in the lab. Further, protein-DNA conjugates were made by attaching protein to the DNA.
Then, the DNA circuits on E.coli bacteria and human cells were tested by the researchers. The powerful DNA technology and proteins within the cells are linked together in this study. They also showed that cancer prodrug, 5-fluorocytosine can be activated into 5-fluorouracil, which is the toxic chemotherapeutic form of 5-fluorocytosine. Particular DNA or RNA sequence inputs will turn on the DNA circuit and protein, and the system is turned off if these inputs are absent. By placing these sequence inputs on microRNA, scientists were able to do this. Apart from the applications of treating cancer, it can also be used in the production of biofuels.
This new technology can be used to target any DNA sequence and attach protein of their choice. According to the researchers, in future, this technology will be able to address a variety of diseases.
